Phase 2, randomised, double-blind, placebo-controlled dose-ranging trial of retatrutide in adults with obesity or overweight and at least one weight-related comorbidity, excluding type 2 diabetes. N=338. This is the original Phase 2 obesity trial and its 48-week results, published in the New England Journal of Medicine, first established retatrutide’s headline efficacy: mean percent change in body weight from baseline at week 48 was -24.2% on the highest studied dose regimen (12mg, titrated via a 2mg starting dose) versus -2.1% on placebo, with clear dose-dependence across the 1mg, 4mg, 8mg and 12mg arms. These are 48-week, mixed-effects-model efficacy-estimand figures from a Phase 2 trial with a comparatively small placebo arm — not the larger, longer Phase 3 TRIUMPH programme data, and not necessarily reproduced at Phase 3 scale.
